Analysis of blood for secnidazole in female volunteers by HPLC method

Secnidazole is an antiprotozoal compound nitroimidazole family prescribed against acute intestinal amoebiasis, giardiasis, bacterial vaginosis and trichomoniasis. The biokinetic parameters were investigated in ten healthy female volunteers after 1gm oral administration of secnidazole. After single oral administration of secnidazole the mean maximum concentration of secnidazole in plasma was 23.08 + 0.80 after 2.5 hours. The area under curve (AUC) of volunteers was 349.05 plus minus 20.9h. mg/L. The value for clearance was 0.44 plus minus 0.31 L/h. The value for volume of distribution was 11.09 plus minus 7.81L. The values for elimination half-life and mean residence time were 13.86 plus minus 0.92h, 20.61 plus minus 1.36h and the values for absorption rate constant absorption half-life were 2.97 plus minus 0.46 L/h and 0.27 plus minus 0.03h respectively. The lag time of the secnidazole was 0.22 plus minus 0.05 hours. The rate constant was 0.05 plus minus 0.002 L /h. The mean time for secnidazole to reach maximum concentration was 1.77 plus minus 0.16h and maximum concentration achieved was 16.24 plus minus 0.48 mg/L. The study supports the need for the comprehensive evaluation of drug under our own environmental conditions to obtain biodisposition kinetic parameters on which the rational dose regimens of drug could be based.
