For asparagus anther culture, 2,4-d played a leading role in stimulating callus ploidy variation. High concentration of 2,4-d increased both callus polyploidy and callus induction rate. The differentiation media with different rates of growth regulators demonstrated distinct difference in inducing ploidy variation for regenerated plants. The frequency of haploids in the shoot tip of the regenerated plant on the medium supplemented with 1.0 ppm BA and 0.2 ppm IBA, was the highest (31.17%). Comparison revealed that the ploidy variation of the regenerated plants was much larger than that of the calli.
