Dopamine and the inhibition of indole-3-acetic acid oxidase in tobacco thin cell layers.

The fate of (l-to the 14th C) indole-3-acetic acid (IAA) added to thin layers of tobacco after 24 hours in culture with and without dopamine addition was investigated. Results show that the presence of dopamine inhibits the breakdown of (l-to the 14th C) IAA by as much as 75 percent in the tobacco explants. Evidence is presented to show that the protective action of dopamine is achieved by inhibiting IAA oxidase, an enzyme believed to be responsible for auxin breakdown. Results also indicate that the exogenous IAA was rapidly conjugated to two major metabolites after 12 hours, one is aspartate, accounting for about 50 percent of the label in dopamine-treated extracts, and the other compound accounting for about a third of the auxin is suspected to be glycine.
