Evaluation of cyclodiene immunoassay kit for residue analysis.

An immunoassay kit for cyclodiene was evaluated for residues in soil and water in terms of reproducibility and matrix effects. Comparison of results was made by using gas liquid chromatography. The absorbance readings using ELISA were inversely proportional to the concentration of dieldrin with an equation of percent B=113.62-31, 41 log C and a correlation coefficient of 0.945 at a concentration range of 5-100 mg/ml. Source of water was found to interfere with absorbance readings with slope of the line greatest in distilled water followed by paddy water and tap water. Dilution of the extracts by itself did not affect absorbance readings but an increase sensitivity with dilution was observed on soil with low organic matter. Dilution did not improve quantitative detection of dieldrin in soil with high organic matter. The sensitivity of the kit using prepared dieldrin standards was high but slope values indicate greater response for change in concentration for the chlordane calibrators. The use of the immunoassay kit was found to provide a semiquantitative screening method, based on gas chromatographic analysis, for cyclodiene residues in soil and water.
