Karyological study of Abies sp. callus culture.

Callus cultures were obtained from intact embryo plus hypocotyl and radicle segment culture of Abies alba and Abies concolor x Abies grandis seedlings. The most appropriate explants were the hypocotyl segments. Karyological analysis of callus showed that callus cells contained predominantly diploid chromosome number. Karyological changes occurred in max. 10.8 per cent of mitotic cells. The most frequent change was polyploidy. Polyploid cells occurred rarely, the tissue showed no tendency toward polyploidization. Other karyological changes occurred only in single cases.
