Geminiviruses are an economically important group for plant viruses characterized by their twinned virions and circular single stranded DNA genomes. Recent evidence indicates that genetic variation exists among geminiviruses infecting beans (Phaseolus vulgaris). Geminiviruses are well suited to detection and identification by DNA based technologies such as DNA hybridizations and polymerase chain reactions (PCR). Dot and squash blot DNA hybridization methods involve simple sample preparation. For dot blots, leaf tissue is ground in water or a buffer and applied to nylon membranes. General probes for the detection of all known whitefly-transmitted bean-infecting geminiviruses are available. These probes are a mixture of DNA-A's of selected bean-infecting geminiviruses. Specific probes (DNA-B's) for each of the bean-infecting geminiviruses are known and can be used to detect only isolates of BGMV type I, BGMV type II, BDMV or BCMoV, and these specific probes do not cross-hybridize.
