A study of natural resistance of several cereal species to antibiotic Kanamycin

The natural ability of wheat (Triticum aestivum) maize (Zea mays) and barley (Hordeum vulgare) to form calli from mature embryos and regenerate on medium containing 50, 100, 150, 300, 500, 800 mg/l kanamycin was studied. After three weeks there was no difference in the number of calli formed on callus initiation medium either with or without kanamycin, but shoot formation was inhibited on regeneration media which contained kanamycin. This suggests that kanamycin has no effect on callus formation in wheat, maize and barley but inhibits the formation of shoot primordia on regeneration medium. This observation may be used in future for selection of transformed cereal tissues.
