[Continual enzymatic hydrolysis of the starch]

The enzymatic hydrolysis of starch has been modelled with normal corn starch and amylopectin corn starch. In one step in continuum the starch is gelatinized and liquefied with steam injection using microbial alfa amylase (TERMAMIL 120L). After liquefaction, the pH is lowered to 4.5 and temperature is dropped to 55 deg C. The limit dextrins in the substrate pass through immobilised glucoamylase in packed bed reactor. High liquid chromatography was used for following the change of carbohydrate composition during the continual saccharification process. Starch in continual enzymatic hydrolysis with bacterial alfa amylase and immobilized glucoamylase in packed bed may obtain DE value (dextrose equivalent) over 96 and glucose content up to 93. Columns packed with continual immobilized glucoamylase offer new production possibilities in biotechnology processes as well as in carbohydrate chemistry.
