Analgesic effect of opioid peptides against visceral pain in rats can be potentiated by neutral endopeptidase inhibitor

The results show that N-carboxymethyl-Phe-Leu (NcPL) icv applied alone exerted significant and dose-dependent analgesic effect. Leu-enkephalin had no analgesic effect, but after inhibition of neutral endopeptidase with NcPL exerted significant analgesic effect. Met-enkephalin, alone and after pretreatment with NcPL exerted significant analgesic effect. The present study demonstrates that Leu-enkephalin, probably is not included in response to chemical painful stumuli in rats, in contrast to Met-enkephalin, but after inhibition of their enzymatic degradation analgesic effect of both can be significantly potentiated.
