Chlorophyll and carotenoid accumulation in cotyledons of Acer pseudoplatanus L. seedlings grown in the light and in the dark

In order to determine the pattern of photosynthetic apparatus transformations in cotyledons of Acer pseudoplatanus seedlings germinated in the light and in the dark the changes of chlorophyll and carotenoid content were studied. In the light the onset of rapid chlorophyll accumulation coincided with the splitting of the seed coat and emergence of the radicle and cotyledons. In the dark at the beginning of germination the chlorophyll content dropped tp half the original value; later four-fold rising suggests that chlorophyll synthesis occurs in the dark. Carotenoid accumulation roughly followed chlorophyll accumulation both in the light and in the dark. Pigment composition was similar to that of the leaves. In the dark, a lack of beta-carotene was noticed. In the light beta-carotene appeared in unfolded cotyledons suggesting that its main function may be protection of chlorophyll from photobleaching.
