DISEÑO Y ELABORACIÓN DE TABLETAS REVESTIDAS DE DARUNAVIR, 800 mg. | DESIGN AND PRODUCTION OF DARUNAVIR COATED TABLETS, 800 mg.

Spanish; Castilian. Darunavir es un fármaco antirretroviral de uso sistémico. Se emplea para el tratamiento de la infección por el Virus de la Inmunodeficiencia Humana de tipo 1 (VIH-1) co-administrado con ritonavir y con otros agentes antirretrovirales. Su uso es bastante común en Cuba, como producto de importación, con un costo significativo. En la actualidad como parte de la estrategia nacional de sustitución de importaciones, el Centro de Investigaciones y Desarrollo de Medicamentos (CIDEM) tuvo como objetivo desarrollar este producto con una formulación y proceso nacional, que posea una excelente calidad y seguridad para el tratamiento de tan grave enfermedad. Durante el desarrollo de la investigación se diseñaron un total de nueve formulaciones, todas elaboradas por el método de granulación húmeda, donde se examinaron la influencia en la calidad de dos agentes aglutinantes, dos excipientes de compresión directa, así como dos solventes y variaciones en el contenido de aglutinantes y lubricantes. La formulación seleccionada como mejor, empleó PVP K-40, en solución alcohólica y Encompress. Con la fórmula seleccionada se elaboraron tres lotes a escala piloto de tabletas oblongas de color azul, revestidas con una película hidrosoluble de Polycoat YS 1- 7003 que demostraron excelente propiedades físico-mecánicas y tecnológicas.

English. Darunavir is a pharmaceutically active ingredient antirretroviral of systemic use. It is used for the treatment of the infection by the Human Inmunodeficiency Virus type 1 (HIV-1) co-administered with ritonavir and with other antirretrovirals agents. Their use is quite common in Cuba, as import product, with a significant cost. At the time, like part of the national strategy of substitution of imports, the Center of Investigations and Development of Medications (CIDEM) had as objective to develop this product with a national formulation and process, an excellent quality and security for the treatment of so serious illness. During the development of the investigation were designed a total of nine formulations, all elaborated by the method of humid granulation, the influence in the quality of two agglutinative agents were examined, also of two exicipients of direct compression, as well as two solvents and variations in content of glidants and lubricants. The formulation selected as better, used PVP K-40, in alcoholic solution and encompress, besides the other exicipients. With the selected formula three lots were elaborated to scale pilot of oblong tablets of blue color, had with a polycoat hidrosoluble YS 1 – 7003 film, they demonstrated they demonstrated excellent physical-mechanical and technological properties.
