Dinámica en la producción y consumo de vitamina B12 entre grupos bacterianos protótrofos y auxótrofos | Dynamics in the production and consumption of vitamin B12 between prototrophic and auxotrophic bacterial groups

La vitamina B12 o cobalamina comprende un conjunto de moléculas orgánicas, esenciales para el crecimiento de diversos organismos eucariotas y procariotas. Esto se debe a que actúa como cofactor de diferentes enzimas involucradas en el metabolismo energético y no energético. En el medio marino la B12 es sintetizada por ciertos organismos procariotas. Sin embargo, poco se sabe sobre qué organismos son capaces de incorporarla a través de la vía de reciclaje o salvamiento. Además, se desconoce cómo la expresión de genes relacionados con la síntesis, dependencia y salvamiento puede ser afectada por la variabilidad temporal en el ecosistema. El presente trabajo busca describir la distribución de la capacidad de síntesis, dependencia y salvamiento de B12 en genomas de los principales grupos taxonómicos del bacterioplancton marino. Asimismo, busca determinar patrones temporales de la expresión de estos genes en una comunidad microbiana marina. Se utilizó la base de datos IMG (Integrated Microbial Genomes) de la cual se extrajeron 1626 genomas de organismos procariotas de origen marino. En estos genomas se identificaron los genes asociados a la ruta de síntesis y enzimas dependientes de B12. Por otro lado, se evaluó la expresión de dichos genes mediante metatranscriptómica en muestras ambientales recolectadas en cuatro estaciones del año en Isla Sapelo, Georgia, EUA. Se determinó que los principales taxones con genomas sintetizadores de B12 pertenecen a Cyanobacteria (78 %), Alphaproteobacteria (38 %) y Gammaproteobacteria (5 %). Más del 90 % de los genomas de cada grupo taxonómico presentaron al menos una enzima dependiente de B12. Además, se determinó que la mayoría (907) no cuenta con la capacidad de síntesis de cobalamina, pero potencialmente pueden reciclar moléculas degradadas de B12 a través de la vía de salvamiento. Por otra parte, se observó que la expresión de genes de síntesis y dependencia de B12 en Isla Sapelo es llevada a cabo por distintos grupos taxonómicos a lo largo del año. En conclusión, nuestro estudio sugiere que organismos que no producen cobalamina dependen de una fuente exógena o vía de salvamiento, y que la expresión de estos genes está influenciada por la variabilidad estacional.

Vitamin B12 or cobalamin comprises a group of essential organic molecules crucial for the growth of various eukaryotic and prokaryotic organisms. This is attributed to its role as a cofactor of different enzymes participating in both energy and non-energy metabolism. It is established that certain marine prokaryotic organisms are responsible for the synthesis of B12 in this environment. However, there is a lack of understanding about the presence of the salvage pathway and how the expression of B12 synthesis, dependence and salvage genes may be affected by temporal variability in the ecosystem. The present thesis seeks to describe the distribution of the synthesis, dependence and salvage of B12 genes in genomes of the main taxonomical groups that are part of the marine bacterioplankton. In addition, this works aims to determine patterns of the expression of these genes in a marine microbial community associated with the temporal variability. The IMG (Integrated Microbial Genomes) database was used, to extract 1626 genomes of prokaryotic organisms from the marine ecosystem. In these genomes, genes associated with B12 synthesis and dependent enzymes were searched for. On the other hand, the expression of these genes was assessed through metatranscriptomics in environmental samples collected during four seasons of the year on Sapelo Island, Georgia, USA. The main taxonomic groups with de novo B12 synthesizing genomes were Cyanobacteria (78 %) and the Alpha- (38 %) and Gammaproteobacteria (5 %) classes. At the same time, more than 90 % of the genomes of each taxonomic group analyzed presented at least one B12-dependent enzyme. Ribonucleotide diphosphate reductase and methionine synthase were B12-dependent enzymes most commonly found. Furthermore, it was determined that some genomes (907) do not synthesize cobalamin, but instead have the genes associated with the salvage pathway. On the other hand, the expression of B12 synthesis and dependence genes on Sapelo Island is carried out by different taxonomic groups throughout the year. In conclusion, our study suggests that some organisms that do not produce cobalamin but depend on an exogenous source or salvage pathway of B12, and the gene expression is influenced by seasonal variability.
