Untersuchung der Protein-DNA-Interaktion am Replikationsursprung in der Lysozym-GAS41-Gendomäne des Huhnes (Gallus gallus, Linnaeus, 1758)

German. Die Initiation der DNA-Replikation erfolgt in Eukayonten an zahlreichen Replikationsursprüngen (Origins), die von dem heterohexameren ORC-Komplex erkannt werden. Bei zahlreichen DNA-Viren wurde gezeigt, dass neben dem ORC-Komplex auch Transkriptionsfaktoren wie Oct-1, NF1 und Sp1 für die Initiation der Virus-Replikation notwendig sind. Im Lysozym-GAS41-Genlocus des Huhns existiert ein Ursprung der bidirektionalen DNA-Replikation (OBR). Der OBR liegt in einem engen, etwa 300 bp umfassenden Bereich zwischen dem Lysozymgen und dem direkt stromabwärts angrenzenden GAS41-Gen. Daher überlagert sich der OBR mit dem GAS41-Promotor. Im Bereich des Lysozym-OBR / GAS41-Promotors kommen ebenfalls Bindemotive für Sp1, Oct-1 und NF1 vor. Es wurde vermutet, dass diese Transkriptionsfaktoren auch bei der Initiation der DNA-Replikation am Lysozym-GAS41-OBR involviert sein könnten. Ziel der vorliegenden Arbeit war es, solche Replikations- und Transkriptionsfaktoren zu identifizieren, die mit dem Lysozym-GAS41-OBR des Huhns interagieren und für die Initiation der DNA-Replikation essentiell sind. Dabei sollte auch die DNA-Bindestelle von ORC2, der 72 kDa-Untereinheit des hexameren Hühner-ORC-Komplexes, im Lysozym-GAS41- OBR identifiziert werden. Um DNA-Bindestellen für Initiationsproteine des Lysozym-GAS41-OBRs zu identifizieren, wurden in vitro Protein-DNA-Interaktionen zwischen DNA-Fragmenten aus dem OBR und GAS41-Promotor und Kernextrakten verschiedener Zelllinien (Huhn, human) untersucht. Aus den Gelretardationsanalysen (EMSA und Immunomobility Shift-Assays) ging hervor, dass die Transkriptionsfaktoren Sp1 und Oct-1 mit spezifischen DNA-Bindemotiven innerhalb des OBR/ GAS41-Promotors interagieren. Sp1 interagiert mit einem als "GC-Box" bezeichneten Bindemotiv (5’-(G/T)GGGCGG(G/A)(G/A)(C/T)-3’), wohingegen Oct-1 mit einer "Octamer-Motiv-ähnlichen" Sequenz (5’-ACGCAAAC-3’) interagiert, welche im Vergleich zum bekannten "Octamer-Motiv" (5’-ATGCAAAT-3’) eine Sequenzabweichung von zwei Basenpaaren aufweist. Eine in vitro-Interaktion zwischen NF1 und einer im OBR befindlichen potentiellen NF1-Bindestelle (5’-TTGGCA-3’) wurde nicht festgestellt. Zudem wurde gezeigt, dass ORC2, die etwa 72 kDa große Untereinheit des Hühner-ORC-Komplexes, in vitro mit der 5’-Hälfte einer im OBR/ GAS41-Promotor befindlichen AT-reichen Sequenz interagiert. Die mutmaßliche ORC2-Bindestelle befindet sich direkt stromaufwärts der identifizierten Oct-1-Bindestelle. Um die Bindung von ORC2, Sp1 und Oct-1 am Chromatin des OBRs in vivo nachzuweisen, wurden Chromatin-Immunpräzipitationen (ChIPs) durchgeführt. Hierfür wurde formaldehydbehandeltes und durch Ultraschall fragmentiertes crosslink-Chromatin aus HD11- und DU249-Hühnerzellen mit spezifischen Antikörpern gegen cORC2, hSp1, hOct-1 und hNF1 immunpräzipitiert. Für die ChIP-Experimente wurde auch ein Antikörper gegen Hühner- ORC2 hergestellt, welcher durch Immunisierung eines Kaninchens mit einem bakteriell exprimierten, rekombinanten 28 kDa-cORC2-Polypeptid produziert wurde. Die immunpräzipitierten Nucleoproteine wurden im Western Blot analysiert. Dabei zeigte sich, dass ORC2 sowohl mit Sp1 als auch mit Oct-1 heteromere Proteinkomplexe bildet. Daraus wurde geschlossen, dass ORC2, Sp1 und Oct-1 in vivo an dicht benachbarten Chromatinregionen binden und eng miteinander assoziiert sein könnten. Die in den ChIP-Experimenten coimmunpräzipitierten Chromatinfragmente wurden mittels quantitativer Real-Time PCR analysiert. Dabei wurde eine signifikante Anreicherung von Sequenzen aus dem OBR/ GAS41-Promotorbereich in den anti-cORC2-, anti-hSp1- und anti-hOct-1-Immunpräzipitaten festgestellt. Daraus wurde geschlossen, dass ORC2, Sp1 und Oct-1 auch in vivo am Chromatin des Lysozym-GAS41-OBRs binden und dass ihre Bindung vermutlich an den in den in vitro-Experimenten identifizierten Bindestellen erfolgt. Desweiteren wurde vemutet, dass ORC2 in Interaktion mit den beiden Transkriptionsfaktoren Sp1 und Oct-1 an den Lysozym-GAS41-OBR in vivo bindet. Das Vorkommen zahlreicher Sp1-Bindestellen im OBR/ GAS41-Promotor wurde als Indiz für eine Involvierung von Sp1 bei der Initiation der Replikation am OBR diskutiert. Um dem nachzugehen wurden Transfektionsexperimente mit Targeting-Plasmiden und der Hühner-B-Lymphozytenzellline DT40 durchgeführt, um die etwa 320 bp umfassende Sp1-Binderegion in beiden Allelen mittels homologer Rekombination zu deletieren. Nach der ersten Transfektionsrunde wurden insgesamt 160 antibiotikumresistente Zellklone isoliert, 51 Klone aus einem Experiment mit Plasmid pSp1-neo und 109 Klone aus drei Experimenten mit Plasmid pSp1-puro. Die chromosomale DNA der Klone wurde im Southern Blot analysiert. Bei keinem Klon wurde eine homologe Rekombinantion festgestellt. Als Grund hierfür wurde vermutet, dass der in den Plasmiden bereits deletierte Sp1-Bindebereich mit rund 300 bp zu groß für die 3’-angrenzende homologe Sequenz war.

English. In eukaryotes, DNA replication initiates at multiple origins of DNA replication which are recognized by the heterohexameric "origin recognition complex" (ORC). For many DNA viruses it has been shown that, besides ORC, transcription factors such as Oct-1, NF1 and Sp1 are required for the initiation of viral replication. In the chicken lysozyme-GAS41 gene locus an "origin of bidirectional DNA replication" (OBR) has been identified. The OBR resides within a narrow 300 bp region between the lysozyme gene and the immediately downstream localized GAS41 gene. Hence, the OBR overlaps with the GAS41-promoter. The region of the OBR/ GAS41-promoter also contains binding motifs for the transcription factors Sp1, Oct-1 and NF1. It was presumed that these transcription factors may also be involved in the initiation of DNA replication at the lysozyme-GAS41-OBR. Aim of this study was to identify such replication and transcription factors which interact with the chicken lysozyme-GAS41-OBR and which are essential for the initiation of DNA replication. The DNA-binding site of ORC2, the 72 kDa subunit of the hexameric chicken ORC, should also be identified in the lysozyme-GAS41-OBR. To identify DNA-binding sites for initiation proteins of the lysozyme-GAS41-OBR in vitro protein-DNA-interactions between DNA-fragments of the OBR and GAS41-promoter and nuclear extracts from different cell lines (chicken, human) were investigated. These gel retardation experiments (EMSA and Immunomobility Shift Assays) demonstrated that transcription factors Sp1 and Oct-1 interact with specific DNA-binding motifs within the OBR/GAS41-promoter. Sp1 interacts with a binding motif, called "GC-box" (5’-(G/T)GGGCGG(G/A)(G/A)(C/T)-3’), while Oct-1 interacts with an octamer motif-like sequence (5’-ACGCAAAC-3’) which shows two mismatches compared to the known "octamer motif" (5’-ATGCAAAT-3’). An in vitro interaction between NF1 and a potential NF1 binding site within the OBR/ GAS41-promoter (5’-TTGGCA-3’) was not observed. Additionally, it was shown that ORC2, the 72 kDa subunit of the chicken ORC, interacts with the 5’-half of an AT-rich sequence within the OBR/ GAS41-promoter in vitro. The presumed ORC2 binding site is located immediately upstream of the identified Oct-1 binding site. To proof the binding of ORC2, Sp1 and Oct-1 at the OBR chromatin in vivo chromatinimmunoprecipitations(ChIPs) were performed. In the experiments, formaldehyde-treated crosslink chromatin from chicken HD11 and DU249 cells was fragmented by sonication and immunoprecipitated with specific antibodies against cORC2, hSp1, hOct-1 and hNF1. Additionally, an antibody against chicken ORC2 was generated for the ChIP experiments which was produced by immunization of a rabbit using a bacterially expressed, recombinant 28 kDa cORC2-polypeptide. The immunoprecipitated nucleoprotein fractions were analysed by Western Blotting. It was shown that ORC2 forms heteromeric protein complexes with both, Sp1 and Oct-1. From these results it was concluded that ORC2, Sp1 and Oct-1 bind to neighbouring chromatin regions in vivo and are probably closely associated with each other. The coimmunoprecipitated chromatin fragments from the ChIP experiments were analysed by quantitative Real-Time PCR. A significant enrichment of sequences from the OBR/ GAS41-promoter was detected in the anti-cORC2-, anti-hSp1- and anti-hOct-1-immunoprecipitates. This indicates that ORC2, Sp1 and Oct-1 also bind to the chromatin of the lysozyme-GAS41-OBR in vivo, most probably to the binding sites which were identified in the in vitro experiments. It is presumable that ORC2 binds in interaction with both transcription factors, Sp1 and Oct-1 to the lysozyme-GAS41-OBR in vivo. The presence of multiple Sp1 binding sites in the OBR/GAS41-promoter could point to an involvement of Sp1 in the initiation of DNA replication at the lysozyme-GAS41-OBR. To follow this up, transfection experiments with targeting plasmids and the chicken B-lymphocytic cell line DT40 were performed to delete the 320 bp Sp1 binding region in both allels by homologous recombination. After the first round of transfection, a total of 160 antibiotic resistent cell clones were isolated, 51 clones from one experiment with plasmid pSp1-neo and 109 clones from three experiments with plasmid pSp1-puro. The chromosomal DNA from these clones was analysed by Southern Blotting. None of the clones showed a homologous recombination. Possibly, the ca. 300 bp encompassing Sp1 binding region which was already deleted in the plasmids, was to broad for the 3’-neighbouring homologous sequence.
