Untersuchung des Einflusses von autochthonen potentiell pathogenen Bakterien und Laktobazillen auf bovine Endometriumszellen in vitro | Evaluation of the influence of autochthonous potentially pathogenic bacteria and lactobacilli on bovine endometrial cells in vitro

Nach dem Abkalben tritt häufig eine bakterielle Einwanderung in den Uterus des Rindes auf, die zu Krankheiten wie Metritis, chronischer oder subklinischer Endometritis führen kann. Dabei stellt das Endometrium die erste physikalische Barriere für eingedrungene Bakterien dar und ist beteiligt an der Antwort des angeborenen Immunsystems durch die Produktion von Zytokinen, um polymorphkernige neutrophile Granulozyten und Makrophagen anzulocken und zu aktivieren. Das Augenmerk der Forschung lag bisher in diesem Zusammenhang auf pathogenen Bakterien, die uterine Erkrankungen beim Rind hervorrufen können. Der Einfluss von kommensalen und potentiell pathogenen Bakterien auf das bovine Endometrium ist dagegen weitgehend unerforscht. Ziel des Forschungsprojektes war es daher, potentiell pathogene Bakterien sowie kommensale Lactobacillus spp. aus dem bovinen Uterus zu isolieren und zu charakterisieren. Die inflammatorische Immunantwort des Endometriums sollte in vitro und für bestimmte Entzündungsfaktoren in vivo untersucht werden. Nicht-trächtigen, gesunden Kühen wurden mittels Cytobrushmethode endometriale Proben entnommen und daraus bakterielle Isolate gewonnen. Im ersten Teil dieser Arbeit wurde das pathogene Potential von den autochthonen Isolaten Bacillus pumilus, Staphylococcus succinus und Weissella paramesenteroides verglichen, um eine erste Immunantwort des bovinen Endometriums auf eine bakterielle Infektion in vitro zu untersuchen. B. pumilus verursachte innerhalb von 24 Stunden den Zelltod kokultivierter Epithelzellen und wurde als der potentiell pathogenste Kandidat für weitere in vitro Versuche ausgewählt. Bovine endometriale Epithelzellen wurden mit B. pumilus in unterschiedlicher Anzahl (Multiplicity of infection (MOI) 1, 5 und 10) für 2, 4 und 6 Stunden kokultiviert. Gesamt-RNA wurde aus den kokultivierten Endometriumszellen isoliert. Mittels quantitativer reverser Transkriptions-Polymerase Kettenreaktion (RT-qPCR) wurde die Quantifizierung der mRNA-Expression der Entzündungsmediatoren Interleukin 1 alpha (IL1A), IL6, IL8, Prostaglandin-Endoperoxid-Synthase 2 (PTGS2), der CXC-Chemokin-Liganden 1-3 (CXCL1-3), des CXC-Chemokinrezeptors 2 (CXCR2), des Toll-like-Rezeptors 2 (TLR2) und TLR6 durchgeführt. Zum Vergleich mit der mRNA-Expression von Entzündungsfaktoren in vivo wurden endometriale Cytobrushproben von gesunden Kühen und Kühen mit subklinischer oder chronischer Endometritis entnommen und mittels RT-qPCR analysiert. Die mRNA-Expression von CXCL1/2 und CXCR2 war in Endometriumsproben von Kühen mit subklinischer Endometritis und von CXCL3 bei Kühen mit chronischer Endometritis verglichen mit Endometriumsproben von gesunden Kühen signifikant erhöht. B. pumilus verursachte vergleichbar zu den in vivo Ergebnissen eine signifikant erhöhte mRNA-Expression von CXCL1-3, CXCR2, IL1A, IL6, IL8 und PTGS2 in den kokultivierten bovinen endometrialen Epithelzellen verglichen mit Kontrollzellen. Die Entzündungsfaktoren zeigten bereits nach zwei Stunden Kokultivierung mit B. pumilus in fast allen MOI die größte Erhöhung der mRNA-Expression im Vergleich zur Kontrolle. Im zweiten Teil dieser Arbeit wurden verschiedene Milchsäurebakterien wie Lactobacillus buchneri, L. ruminis, L. amylovorus, L. plantarum, L. similis, Weissella paramesenteroides und Pediococcus pentosaceus aus endometrialen Proben nicht-trächtiger, gesunder Kühe identifiziert. Die autochthonen Isolate L. buchneri, L. ruminis und L. amylovorus und ein kommerziell erhältlicher L. vaginalis wurden in einer MOI von 1, 5 und 10 mit bovinen endometrialen Epithelzellen kokultiviert. Die Laktobazillen zeigten keinen negativen Einfluss auf die Vitalität der kokultivierten Epithelzellen. Mittels RT-qPCR wurde die Quantifizierung der mRNA-Expression von IL1A, IL6, IL8, PTGS2, TLR2 und TLR6 nach 2, 4 und 6 Stunden durchgeführt. Die Freisetzung von IL6, IL8, Prostaglandin E2 (PGE2) und PGF2α wurde mittels Enzyme-linked Immunosorbent Assay (ELISA) oder Enzyme Immunoassay (EIA) nach 24 und 48 Stunden bestimmt. L. ruminis und L. amylovorus bewirkten in den Epithelzellen eine gesteigerte mRNA-Expression von IL1A, IL6, IL8 und PTGS2 sowie eine erhöhte Freisetzung von IL8 und PGF2α im Vergleich zu Kontrollzellen. Im Gegensatz dazu beeinflusste L. buchneri die Expression und Freisetzung der untersuchten Faktoren nicht signifikant. Die mRNA-Expression von TLR2 und TLR6 zeigte sich nach der Kokultivierung unverändert zu den Kontrollzellen. Die Ergebnisse der beiden Studien legen den Schluss nahe, dass sowohl potentiell pathogene Bakterien als auch bestimmte Kommensale die angeborene Immunantwort in bovinen endometrialen Epithelzellen auslösen können. Die Feststellung, dass L. buchneri keine Immunantwort in kokultivierten endometrialen Epithelzellen auslöste, könnte in der Prophylaxe oder Therapie von uterinen Erkrankungen Verwendung finden.

Bacterial invasion of the uterus, which is able to lead to diseases like metritis, clinical and subclinical endometritis, is common in cattle after calving. The endometrium represents the first physical barrier for invading bacteria and is involved in the innate immune response by producing cytokines to attract and activate polymorphonuclear neutrophil granulocytes and macrophages. So far, the focus of research in this context has been on pathogenic bacteria that can cause uterine bovine diseases. However, the influence of commensal and potentially pathogenic bacteria on the bovine endometrium is mainly unknown. Therefore, the aim of the research project was to isolate and characterize potentially pathogenic bacteria and commensal Lactobacillus spp. from the bovine uterus. The inflammatory immune response of the endometrium was examined in vitro and for selected inflammatory factors in vivo. Endometrial samples were collected from non-pregnant healthy cows by using the cytobrush method and out of them bacterial isolates were gained. In the first part of this project, the pathogenic potential of the autochthonous isolates Bacillus pumilus, Staphylococcus succinus, and Weissella paramesenteroides was examined to reveal a first immune response to a bacterial infection of the bovine endometrium in vitro. B. pumilus caused cell death of co-cultured epithelial cells within 24 h and was selected as the potentially most pathogenic candidate for further in vitro experiments. Bovine endometrial epithelial cells were co-cultured with B. pumilus at a multiplicity of infection (MOI) of 1, 5, and 10 for 2, 4 and 6 h. Total RNA was isolated from co-cultured endometrial cells and subjected to quantitative reverse transcription-polymerase chain reaction (RT-qPCR) for quantification of the mRNA expression of the inflammatory mediators interleukin 1 alpha (IL1A), IL6, IL8, prostaglandin endoperoxide synthase 2 (PTGS2), CXC chemokine ligands 1-3 (CXCL1-3), CXC chemokine receptor 2 (CXCR2), Toll-like receptor 2 (TLR2) and TLR6. Endometrial cytobrush samples from healthy cows and cows with subclinical or clinical endometritis were collected for comparison with the mRNA expression of inflammatory factors in vivo and analyzed by using RT-qPCR. CXCL1/2 and CXCR2 mRNA expression was significantly increased in endometrial samples from cows with subclinical endometritis and CXCL3 in cows with clinical endometritis compared to endometrial samples from healthy cows. Comparable to the in vivo results, B. pumilus caused a significantly increased mRNA expression of CXCL1-3, CXCR2, IL1A, IL6, IL8, and PTGS2 in co-cultured bovine endometrial epithelial cells compared to control cells. The inflammatory factors showed the highest increase of mRNA expression in almost all MOI after only two hours of co-cultivation with B. pumilus compared to the control. In the second part of this work, various lactic acid bacteria such as Lactobacillus buchneri, L. ruminis, L. amylovorus, L. plantarum, L. similis, Weissella paramesenteroides and Pediococcus pentosaceus were identified from endometrial samples from non-pregnant healthy cows. The autochthonous isolates L. buchneri, L. ruminis and L. amylovorus and a commercially available L. vaginalis were co-cultured with bovine endometrial epithelial cells at a MOI of 1, 5, and 10. The lactobacilli showed no negative impact on the viability of the co-cultured epithelial cells. By using RT-qPCR, quantification of the mRNA expression of IL1A, IL6, IL8, PTGS2, TLR2 and TLR6 was performed after 2, 4, and 6 h. The release of IL6, IL8, prostaglandin E2 (PGE2) and PGF2α was measured by using enzyme-linked immunosorbent assay (ELISA) or enzyme immunoassay (EIA) after 24 h and 48 h. L. ruminis and L. amylovorus caused in the epithelial cells increased mRNA expression of IL1A, IL6, IL8, and PTGS2 as well as a higher release of IL8 and PGF2α compared to control cells. In contrast, L. buchneri did not significantly affect the expression and release of the examined factors. The mRNA expression of TLR2 and TLR6 was unaffected after co-culture compared to the control cells. The results of the two studies suggest that potentially pathogenic bacteria as well as distinct commensals may induce an innate immune response in bovine endometrial epithelial cells. The finding that L. buchneri elicits no immune response in co-cultured endometrial epithelial cells could be used in the prophylaxis or therapy of uterine diseases.
