Immunrespons mot Pasteurella spp. hjå rognkjeks (Cyclopterus lumpus L.) og Atlantisk laks (Salmo salar L.) | Immune response against Pasteurella spp. in lumpfish (Cyclopterus lumpus L.) and Atlantic salmon (Salmo salar L.)

Dei siste åra har ein sett ei dramatisk auke i tilfelle pasteurellose i norsk oppdrett av rognkjeks (Cyclopterus lumpus L.) og Atlantisk laks (Salmo salar L.). Sjukdommen førar til redusert fiskevelferd og kan innebere høg dødelegheit. Isolata av Pasteurella spp. som gjev sjukdom hjå rognkjeks og laks er nært i slekt, men likevel ulike. Veterinærinstituttet har føreslått arbeidsnamna Pasteurella atlantica genomovar cyclopteri og Pasteurella atlantica genomovar salmonicida for isolata henta frå henhaldsvis rognkjeks og laks. Om ulikskapane funnen på genomnivå òg har noko å seie for virulens og immunogenisitet veit ein ikkje enno. Kunnskap om fiskane sine immunresponsar mot bakteriane kan nyttast for å finne førebyggjande tiltak og for å utvikle vaksiner, som er viktig for å få kontroll på sjukdommen. Aktivering av leukocyttar, fagocytose og intracellulært drap er viktige prosessar som initiellt må setjast i gang for å oppnå ein beskyttande immunitet mot eit patogen. Fagocytterande leukocyttar tar opp infiserande patogen og brukar både oksygenavhengige og oksygenuavhengige mekanismar for å bryte ned patogenet. Ved oksygenavhengig nedbryting produserer leukocytten reaktive oksygenstoff (ROS) i ein prosess kalla respiratory burst. Hensikta med denne oppgåva var å karakterisere immunresponsen mot Pasteurella spp. in vitro i leukocyttar frå rognkjeks og laks, ved å undersøkje respiratory burst aktiviteten til leukocyttane, samt undersøkje om serum frå rognkjeks immunisert med P. atlantica genomovar cyclopteri kryssreagerar med protein hjå P. atlantica genomovar salmonicida. Dette kan bidra til å påvise antigene likskapar, noko som har betyding for utvikling av vaksinar og fiskens evne til å ha brei eller spesifikk beskyttelse mot ulike isolat. Respiratory burst aktivitet mot to isolat av P. atlantica genomovar cyclopteri og tre isolat av P. atlantica genomovar salmonicida blei undersøkt. Både rognkjeks- og lakseleukocyttar viste respiratory burst aktivitet ved bakteriestimulering, men i ulik grad for dei ulike isolata. Eitt P. atlantica genomovar cyclopteri isolat skilde seg ut ved at det stimulerte til signifikant høgare respiratory burst aktivitet enn den til ustimulerte leukocyttar, frå både rognkjeks og laks. Dette tydar på at det er variasjon i virulens hjå ulike isolat av same bakterie. Serologiske analysar som western blot med serum frå rognkjeks immunisert med P. atlantica genomovar cyclopteri viste tilstadeverelse av kryssreagerande antistoff mot protein hjå P. atlantica genomovar salmonicida. Dette betyr at det er antigene likskapar, men det var òg immunreaktive band som var særskilte for isolata.

In recent years there has been a dramatic increase in reported cases of pasteurellosis in Norwegian aquaculture of lumpfish (Cyclopterus lumpus L.) and Atlantic salmon (Salmo salar L.). Reduced fish welfare and high mortalities are some of the consequences caused by the disease. The isolates of Pasteurella spp. causing disease in lumpfish and salmon are closely related but still different. The Norwegian Veterinary Institute have suggested the working nomenclature Pasteurella atlantica genomovar cyclopteri and Pasteurella atlantica genomovar salmonicida for isolates retrieved from lumpfish and salmon respectively. If the differences found on a genomic level have implications on virulence and immunogenicity is not known. Knowledge about the fish immune responses against these bacteria can support the identification of prophylactic measures and the development of vaccines, which are important measures to control the disease. Activation of leucocytes, phagocytosis and intracellular killing are important processes that initially needs to be started to acquire a protective immunity against a pathogen. Phagocytic leucocytes engulf infective pathogens and use both oxygen-dependent and oxygen-independent mechanisms to break down the pathogen. By oxygen-dependent degradation, the leucocyte will produce reactive oxygen species (ROS) in a process called respiratory burst. The purpose of this study was to characterize the immune response against Pasteurella spp. in vitro using leucocytes from lumpfish and Atlantic salmon, by analyzing the respiratory burst activity of the leucocytes and the cross reactivity of serum harvested from lumpfish immunized with P. atlantica genomovar cyclopteri against proteins from P. atlantica genomovar salmonicida. This can contribute to identify antigenic similarities, which will be important for the development of vaccines and the ability of the fish to have broad or specific protection against different isolates. Respiratory burst activity against two isolates of P. atlantica genomovar cyclopteri and three isolates of P. atlantica genomovar salmonicida were investigated. Both lumpfish and salmon leucocytes showed respiratory burst activity in response to bacterial exposure, but in varying degrees. One P. atlantica genomovar cyclopteri isolate stood out by stimulating both lumpfish and salmon leucocytes to significantly higher respiratory burst activity than unstimulated leucocytes, indicating that there are differences in virulence among different isolates of the same bacteria. Serological analyses by western blot using serum from lumpfish immunized with P. atlantica genomovar cyclopteri showed the presence of cross-reacting antibodies to proteins from P. atlantica genomovar salmonicida. This means that there are antigenic similarities, but there were also immuneoreactive bands that were specific for the isolates.
