Atšķirīgu barotņu ietekme uz krūts vēža šūnu līniju fenotipiskām pazīmēm | Effect of different culture media on the phenotype of breast cancer cell lines

Krūts vēzis ir ļaundabīgs audzējs, kas atšķiras gan viena audzēja ietvaros, gan starp indivīdiem. Lai noskaidrotu pacientam piemērotāko ārstēšanas veidu, tiek noteikti vēža tipi pēc tos veidojošo šūnu morfoloģijas, producētajiem receptoriem, kā arī gēnu ekspresijas. Šūnu kultūrās barotņu sastāvs un citi augšanas apstākļi ietekmē kultivēto šūnu signālprocesus un gēnu ekspresiju. Darbā tika pārbaudīta četru atšķirīgu barotņu ietekme uz krūts vēža līniju (MCF7 un MDA-MB-231) un fibroblastu (HDFa) šūnu līnijas augšanu un fenotipu. Atšķirības tika konstatētas, imūnfluorescenti salīdzinot proteīnu ekspresijas intensitāti, izmantojot augstas caurlaidspējas attēlieguves sistēmu InCell analyzer 2200. Atšķirīgos barotņu sastāvos, izmainījās šūnu veidotie proteīni un fenotips.

Breast cancer is malignant tumor with high heterogenity between individual patients and even within one tumor. Cancer subtypes are classified by cell morphology, produced receptors and gene expression. This information is necessary for choosing the most appropriate therapy. Components of cell culture media and other factors affect cell signalling and gene expression. Therefore, the aim of this study is to compare three different cell lines cultured in four types of media by using immunofluorescence, observe changes in growth and phenotype. The expression of proteins in breast cancer cell lines (MCF7 and MDA-MB-231) and stromal fibroblast cell line HDFa was compared by using InCell analyzer 2200 imaging system. The protein expression and cell phenotype changed in different cell culture media.
