Propagation of Gypsophila paniculata L. [in Family Caryophyllaceae] through tissue culture

Shoot tips, leaf discs, and internodes of Gypsophila paniculata L. were cultured on modified Murashige and Skoog (1962) (MS) and Vacin and Went (1949) (VW) media containing 0.05 to 4 mg/1 NAA. Leaf discs and internodes produced callus at the most rapid rate in MS basal and modified VW media without growth regulator. The most satisfactory callus production was obtained in MS media supplemented with 2 mg/1 NAA or 0.5 mg/1 2, 4-D. The shoot differentiation was induced by using MS media containing 0.5 to 1 mg/1 NAA and 1 to 4 mg/1 BA. Shoot multiplication was obtained on MS and VW media containing various concentration of NAA and BA. The number of shoot and the growth of the multiple shoots grown on a modified VW with double salt strength and 0.5 mg/1 BA were the best and had more desirable morphological characteristics than those grown on the other media. Transplanting hardened plantlets onto a medium consisting of coarse sand and rice hull charcoal (1:1) was very successful. The survival rate was as high as 90 percent.
