Pulmonary cedema and emphysema in buffaloes induced by intravenous administration of 3-Methylindole

Pulmonary oedema and emphysema was induced in buffalo-calves (Bubalus bubalis) by intravenous administration of 3-Methylindole (3MI) 0.06 g/kg body weight. The animals developed acute pulmonary oedema and emphysema, and had average survival time of 60 hr after the infusion. Buffalo calves which received the same dose of 3MI during 2 hrs infusion had no mortality till 168 hr. The disease was clinically characterized by severe dysponea, excessive nasal discharge, high rate of respiration, pyrexia and ataxia. The average maximum plasma levels of 3MI in animals given infusion for 3 hrs was 21.0 mg/ml during the third hour as against 15.0 mg/ml in animals which received the same dose during 2 hrs. On necropsy, the lungs were large, firm, dark red and heavier than normal. Diffuse pulmonary oedema was predominant change in the animals given MI in 3 hr duration. Most alveolar spaces were filled with proteinaceous material, but the alveolar septa were smooth and of normal thickness. In animals given 3MI in 2-hrs duration severe emphysema, proliferation of alveolar cells and thickened alveolar septa were observed
