Studies on the use of thermotherapy and chemotherapy in combination with tissue culture to control cymbidium mosaic virus in Dendrodium pompadour

Preliminary test on inhibition effect of 5 chemicals; 2-thiouracil, 2,4-dichlorophenoxyacetic acid, aspirin, actinomycin D and Hydrotonic, on CYMV infected Den. Pompadour were carried out. The result indicated that 2x10 to -4, 1x10 to -3, 5x10 to -3 M 2-thiouracil and 1.3x10 to -4, 6.8x10 to -4 M 2,4-dichlorophenoxyacetic acid reduced the virus titer as reflected by the reduction in number of local lesion on indicator plants. 1x10 to -3 M 2-Thiouracil and 0.02 percent aspirin injected into the host leaves before inoculation substantially decreased sap infectivity. 1x10 to -3-5x10 to -3 M 2-Thiouracil or 0.0125-0.025 percent Hyrotonic sprayed daily for 15 or 30 days on the infected Den. Pompadour suppressed the virus titer. Virus free shoots were obtained through culturing of shoot meristem, 0.5 and 1 mm. long, on the medium mixed with 0.01, 0.05 and 0.1 percent Hdryotonic but none were found when 2 mm. shoots were used. Culturing of shoot meristem on medium without hydrotonic produced the same result. Immune electron microscope (IEM) and Enzyme-linked immunosorbent assay (ELISA), the serological testmethods, gave reliable results in detecting CYMV infected Den Pompadour
