Genomic libraries for two varieties of cotton (Gossypium hirsutum) were constructed following the methods described by Maniatis and others. One of the two varieties is fusarium wilt-resistant and other sensitive. The numbers of bacteriophage recombinants obtained for the two varieties were calculated at 600000 pfu and 700000 pfu respectively, both exceeding the nesessary number of recombinants that represents an entire genome of cotton according to Clarke and Carbon formula.
