Effect of some culture conditions on phytase biosynthesis by Aspergillus flavipes and Macrophonia phaseolina

This investigation has been adopted to determine the most favourable conditions conducive to the maximum production of both extra and intracellular phytase by Aspergillus flavipes and Macrophomina phaseolina. The data recorded showed that A. flavipes and M. phaseolina produced the highest yields of exo and endeonzyme, resprectively, at pH 6 and 28 C. The two organisms behaved differently toward phytase biosynthesis in presence of various carbon, nitrogen and phosphorus sources. A. flavipes could produce the maximal yield of the extracellutar phytase, amounting to 28.8 units/100 ml, in a medium containing sourose, 5%; (NH4)2 SO4, O,=.3% and Na2 H PO4. 12 H2O, O.O5% after 6 days of incubation. On the other hand, M. phaseolina was able to synthesize 2.28 unites/100 ml of the endoenzyme in presence of starch, 5%; NH4 cl, O.3% and KH2 PO4, O.O3% at the 8th day of incubation. It is of interest to note that C/P ratio plays an important role in phytase production either extra or intracellulary.
