A method for determining apigenin and apigenin 7-0-beta-glucoside in the ligulate flowers of Matricaria chamomilla by applying the combined chromatography-spectrophotometry method, has been developed in this study. Diluted solutions of ethanol are suitable for extracting flavonoids from the ligulate flowers of Matricaria chamomilla. The chloroform-methanol (9:1) solvent system is suitable for separating apigenin, and methylenechloride-methanol (8:2) with 0.1% water for separating apigenin 7-0-beta-glucoside. The concentration of the studied flavonoids in alcohol and water solutions and extracts can be determined with certainty by using UV spectrophotometry. It was found that the ligulate flowers of Matricaria chamomilla contain apigenin (0.33%) and apigenin 7-0-beta-glucoside (2.39%).
