Isolation and characterization of lipolytic bacteria

About 100 samples were tested for isolation of lipolytic bacteria. Primary screening was carried out for prolipase (esterase) activity using tributyrin agar medium followed by confirmation of lipase activity in an olive-oil-agar medium. From 24 isolates with esterase activity, 4 isolates hydrolysed olive- oil resulting in a clear zone formation in the agar medium. These were considered "true" lipolytic bacteria. These 4 isolates were assessed for their lipolytic activity in shake-cultures. One isolate designated as S-20 showed greater lipolytic potentials. This isolate was identified as Pseudomonas spp.
