Propagation in vitro of Aspen Hybrid populus Spartiatica x Populus tremula from mature trees

In Vitro techniques were employed for rapid propagation of four difficult-to-root clones of the aspen hybrid Populus spartiatica x Populus tremula. The initial explants were stem pieces, 1,0-1,5 cm long, consisting of the nodal part and the auxiliary bud and were cut from root suckers of 11-year-old trees. The explants were established on a modified Murashige and Skoog (MS) medium supplemented with 1mg/l benzylaminopurine (BAP), 0,1 mg/l gibberellic acid (GA) and 0,1 mg/1 indole-3-butyric acid (IBA). In the multiplication stage a concentration of 0,2 mg/1 BAP increased the number of shoots which were formed. Six to seven new shoots were harvested in each passage every 21 days. In addition, shoot formation was favoured with 30 g/1 of sucrose in the medium. Differences among the four clones were found on their ability to form shoots in vitro. Shoots harvested form in vitro cultures rooted readily (100% rooting) on a hormone-free medium. Such rooted shoots were transplanted succesfully to a peat: perlite mixture (1:1, V/V) and after a hardening procedure grew normally to plantlets with phenotypic characteristics similar to their parents. Results of the present study indicate that this technigue (utilizing explants from root suckers) will play a decisive role in clonal propagation of aspen hybrids from mature trees.
