Effect of seminal plasma on motility and acrosome morphology of boar spermatozoa stored at 4 degree centigrade

This study was conducted to examine the effect of the seminal plasma on the motility and acrosome morphology of boar spermatozoa stored at 4 degrees C. The sperm-rich fraction of the ejaculates collected from 4 Landrace boars was centrifuged at room temperature and resulting sperm pellets were resuspended in Tris-citric acid-glucose (TCG) solution or in their own plasma. The sperm suspensions were diluted 1:4 (v/v) with egg yolk-TCG (ETCG) diluents containing various concentrations of egg yolk (0, 5, 10, 20% v/v) and catalase (0, 75, 150, 300, 600, 1200 Sigma U./ml), and stored at 4 degrees C for 10 or 15 days. At intervals aliquots were taken and examined. Agglutination of egg yolk was observed microscopically in the samples where sperm suspensions with the plasma had been diluted with diluents containing egg yolk. The presence of the plasma did not affect sperm motility and acrosome morphology for the first 5 days of storage. After 10 days of storage, however, the percentage of progressively motile spermatozoa was higher in the presence of the plasma than in its absence, whereas the percentage of spermatozoa with normal acrosome markedly decreased in its presence. All concentrations of catalase and egg yolk improved sperm motility and acrosomal integrity irrespective of whether the plasma was present or not. The results suggest that there is no need to remove the plasma when semen samples are to be stored at 4 degrees C for 5 days or less. However, if a longer storage period is required, removal of the plasma should be made before dilution
