Some factors affecting callus formation and other responses of kawayan tinik (Bambusa blumeana Schultes f.)

Results showed that there was no significant difference among the three pre-sterilization treatments as to their effect on controlling contamination. However, with their effect on callus formation, phisohex-treated explants yielded the highest number of callus formed followed by those pre-treated with citric/ascorbic acid while the least were those pre-treated with alcohol. Furthermore, those pre-treated with phisohex had significantly less browning than those pre-treated with citric/ascorbic acid. The highest browning occurrence was found on explants pre-treated with alcohol. For the floret explants, it was found that 2% calcium hypochlorite was the most effective in controlling contamination. Among the four media evaluated for leaf explants, namely; Murashige and Skoog's (MS), Woody Plant Medium (WPM), N6 and B5, only MS, WPM had callus formation while those explants in N6 and B5 turned brown with very minimal or no callus at all. Between MS and WPM, MS was found to be more appropriate having produced the most explants with callus. Browning was significantly higher in MS compared to WPM. However, callus grown was still pronounced in MS as evidenced by the development of many profuse callus (D3) type. With floret explants, MS and WPM and N6 and their interaction with light conditions, namely, continuous light, 9/15 hrs. light-dark period and continuous darkness greatly affected callus formation and even callus browning. Explants browning was solely influenced by media. Although MS had the highest explants browning, it produced the most number of profuse callus (D3) type followed by WPM and N6.
