Evaluation of tissue culture in triticale breeding

Anther culture was used to produce haploid plants in 4 hexaploid segregating populations, 1 hexaploid cultivar, 4 F1 octo/hexa crosses, and 3 octoploid lines. The potato 2 (B2) and 4 modifications of B2 medium were tested to induce production of embryogenic calluses (EC). The modified media were: B2 + gln; B2 + gln + casein; B2 + casein + NH4NO3; and B2 + gln + casein + NH4NO3. All media produced EC and no differences could be detected (P less than 0.05) among them. After transference into the regeneration medium, a small number of plants were obtained (140), most of them albinos (112), in comparison to the high number of EC (4338). Five genotypes produced only albino plants, including the one that produced more plants (37). The frequency of albino plants increased and the regeneration capacity decreased with age of the transferred EC. From 28 green plants, 6 were aneuploids and 2 showed spontaneous duplication. After the colchicine treatment 8 duplicated plants produced seeds. At present, the double-haploid plants are being evaluated and modified regeneration media are being tested to increase plant regeneration
