Characterization of glycoconjugates in cloacal epithelium of the chicken

Glycoconjugates in cloacal epithelium of the chicken were studied by means of light microscopic methods of histochemistry. Fixation was performed by immersion with either Carnoy's fluid for 6 hours at room temperature or in Rossman's fluid for 12 hours at 4 deg. Celsius. A variety of staining procedures was applied to 3 um. paraplast sections. These included alcian blue (AB) pH 1.0, AB pH 2.5, periodic acid-Schiff (PAS), AB pH 2.5-PAS, peanut agglutinin (PNA), wheat germ agglutinin (WGA), Dolichos biflorus agglutinin (DBA), Ulex europaeus agglutinin-I (UEA-I) and Limax Flavus agglutinin (LFA) in combination with enzyme digestion with neuraminidase. The free surface at mucosa of coprodeum is covered by simple columnar epithelium in which 2 types of cells can be distinguished-columnar cells, and goblet cells. All mucous granules of goblet cells contain a mixture of acideic and neutral glycoconjugates with sulfate, vicinal diol groupings, N-acetyl-D-glucosamine and sialic acid-galactose dimers. Terminal galactose-(1-3) N-acetylgalactosamine disacchariole was detected in columnar cells of coprodeum. Tubular gland and columnar cells of proctodeum disclosed lectin affinity consistent with the presence of sialic acid-galactose dimers together with terminal galactose-(1-3) N-acetylgalactosamine disaccharide.
