In vitro organogenesis of chickpea and its transformation by Agrobacterium tumefaciens [in Pakistan]

Hypocotyl explants from seedlings of various ages of chickpea were cultured on MS and B5 media supplemented with different combinations of phytohormones. Significant variation was observed among the treatments. MS medium was found to be superior than B5. The best hormonal combination for shoot formation was 5 M benzyladenine (BA) and 2 M indole acetic acid (IAA). Hypocotyl explant from 7-day-old seedlings proved to be the optimal source for culture. Rooting of in vitro regenerated shoot was made in half strength MS medium with 1 M indole-3-butyric acid (IBA). When stems of 10-day.7-old seedlings were infected with a wild-type A. tumefaciens strain (C58), tumors were produced. The induced tumors displayed hormone autonomous growth and were opine positive.
