Preservation methods and their effect on virulence of Pseudomonas syzygii

Nine isolates of Pseudomonas syzygii revived from agar slopes and lyophilized ampules and two unselected fresh field isolates were tested for their virulence on one year old clove seedlings. This experiment showed that isolates of P. syzygii changed their virulence under preservation conditions. Both unselected fresh field isolates were highly virulent indicating as a possible reason for a reported loss of virulence and two isolates revived from lyophil ampules after three years in a storage were also still virulent and killed plants. Only one isolate originally preserved on agar slope showed some ability to invade plants without killing them and all other seven isolates stored on agar slopes were avirulent. The results indicated that lyophilization could be recommended for preserving P. syzygii with little effect on its virulence.
