Effects of pH of culture medium and the temperature during transportation of the ovary on the in vitro fertilization of bovine oocyte

A total of 122 ovaries were collected as aseptically as possible from 61 cows slaughtered. The present study investigated effects of temperature during the transportation of the ovary and the pH of the maturation medium on in vitro fertilization of oocytes. The results obtained were as follows: 1) the maturation medium with pH 7.8 resulted in no development of embryo, while that with pH 7.4 yielded 132 cleaved oocytes from 57 ovaries. 2) Changes in temperature during ovary transportation showed no influence on oocyte maturation, but the temperature range of 31-33 degrees C gave the best result for the penetration of sperms, fertilization and development of oocytes among the tested ranges of 31-33, 34-36 and 37-39 degrees C
