As to cytogenetical studies on variation of Cryptomeria japonica D. Don, karyotype, needle type and stomata on abiogenesis bud mutant and its mother stock were compared. C. japonica clones, Ayasugi (cultivar in Oita district), were used in this experiment. The chromosome number of mother stock observed was 2n=22, and the karyotype was type 0, having F chromosomes without a secondary constriction. The chromosome number of bud mutant observed was 2n=23 (2X+1), trisomics, and H chromosomes were trisomy consisting of metacentric chromosomes. The other chromosomes showed karyotype of type 0, having F chromosomes without a secondary constriction. The typical karyotype of C. japonica such as large trabants (satellites), connecting fiber and Kopfchen (short arm like nipple form) on the J chromosomes was observed in bud mutant and mother stock. Their karyotypes were determined as follows: Mother stock K(22)=2Am+2Bm+2Cm+2Cm+2Dm+2Em+2Fm+2Gm+2Hm+2Im+2scJsm+2Km Bud mutant K(23)=2Am+2Bm+2Cm+2Dm+2Em+2Fm+2Gm+3Hm+2Im+2scJsm+2Km There was no remarkable difference between mother stock (2X) and bud mutant (2X+1) in needle type. However, the depreciate of growth was observed in bud mutant (2X+1). There was no difference also between mother stock (2X) in size of stomata. However, the number of stomata per unit area (0.6*0.4mm) of bud mutant (2X+1) showed decreasing tendency in comparison with mother stock (2X)
