Influences of the sexuality in flower stalk explants on the callus formation and growth in spinach (Spinacia oleracea L.)

In the previous paper, the author reported on the factors affecting callus formation and growth from the explant of young seedling in spinach. The present paper concerned on the differences in callus formation and growth from explants of male and female flower stalkes in spinach. The results obtained may be summarized as follows: 1) Callus formation was observed on the MS media supplemented with auxin, and the best callus formation and growth was obtained on l mM for 2,4-D, on 10 mM for IAA and NAA. 2) Callus formation was observed on the MS media supplemented with cytokinin, and the best results were obtained on 10 mM regardless of kinds of cytokinins. However, the callus growth no media supplemented with cytokinin was inferior than media supplemented with auxins. 3) Callus growth was remarkably promoted on the media supplemented with the most suitable concentrations of auxins and cytokinins. Thidiazuron was most effective for callus growth, following zeatin, BA, 2iP and kinetin. 4) No callus formation was observed on the media supplemented with gibberellin only Gibberellin promoted callus growth on the media supplemented with auxins, but didn't do it on the media supplemented with cytokinins. 5) The minimum concentration of auxins and cytokinins for callus formation was higher at female explants than male. However, the callus growth was better at male than female, especially on the media supplemented with auxins and gibberellin. 6) The above results provide a useful information for understanding the differences between the growth characteristics in sexuality of plant, and for development of micropropagation methods of male and female plants in spinach
