Recovery of oocytes from pig ovarian tissue after puncture of visible ovarian follicles using ovaries obtained from a slaughterhouse

The present study was conducted to investigate the methods for recovery of oocytes from pig ovarian tissue after puncture of visible ovarian follicle using ovaries obtained from slaughterhouse. The pig ovaries were assigned to four treatments: I-II) The surface tissue of ovary was mince by 3 parallel razors at an interval of 1 mm (I) or 2mm (II); and III-IV) The surface tissue of ovary was grated by raddish-grater with large (III) or small (IV) edge. The mean number of oocytes recovered per ovary were 156.6 +-28.6 (I) and 117.6 +- 23.6 (II) by razors and 36.6 +- 10.4 (III) and 50.9 +- 8.9 (IV) by raddish grater, respectively. There was a statistically significant difference between razors groups and raddish-grater groups. But using interval of razors and edge of grater had not influence the number of oocytes recovered per ovary. The recovered oocytes were classified into 6 types based on their morphology in relation to the zona pellucida and surrounding cumulus cells. Treatment method did not influence the rate of oocyte in each rank. But the oocytes surrounded by thick and tight cumulus cell mass formed only 1-8% of total recovered oocytes.
