Leghaemoglobin (Lb) production in pole sitao (Vigna sinensis) and winged bean (Psophocarpus tetragonolobus)

Leghaemoglobin (Lb) contents of the root nodules of pole sitao cv UPL PS1 and winged bean cv Batangas Medium grown in seed pack growth pouches (SPGP) were determined using pyridine haemochromogen method and the Lb solution obtained was applied to disk-gel electrophoresis to determine its components. The gels were scanned with a densitometer at 410 nm. The optical spectra of each Lb band obtained from the densitometric trace was taken by scanning the gel from 400-600 nm. The acetylene reduction activity (ARA) of the root nodules were taken before Lb analysis. Lb production increased with time to about 25-30 percent from 14-21 days after inoculation (dai) in pole sitao and 28-35 dai in winged bean. At 21 dai, Lb contents of pole sitao is 217 nm/g root nodules while in winged beans, 145.52 nm/g at 35 dai. An increase from 25-30 percent caused an abrupt increase from 60-80 percent in ARA. During the later part of winged bean growth, ARA diminished but a corresponding decrease in Lb content was not shown.
