Rapid and quantitative determination of amylose in corn

Authentic amylose and amylopectin solutions (5, 10, 20, 25 and 50 microg starch/ml) with different amylose content (0, 10, 20, 50, 75, 90 and 100 % amy) were stained with J2-KJ solution and absorbancies at two wavelenghts 620 nm (absorption max. for amylose) and 520 nm (absorption max. for amylopectin) were measured. The amylose content of 10 samples of corn was determined according to two evaluated equations and compared with the results found by colorimetric method. It was established the results of both methods are comparable.
