The effect of non-ionic osmotic stress on bean callus cultures

Callus cultures of 26 varieties of bean (Phaseolus vulgaris L.) were maintained on media containing various concentration of mannitol. The induced non-ionic osmotic stress inhibited growth, the relative growth rate is lower and dry matter content is higher in stress sensitive calli than in less sensitive one. Osmotic stress also causes changes in the amino acid and polyamine synthesis. Osmotic treatments increased total free amino acid contents in sensitive bean calli only. In calli of stress sensitive bean varieties arginine, alanine, valine, asparagine acid, threonine, phenylalanine and histidine content increased more than in less sensitive type
