Isolation of amplified DNA from microdissected 5BLdel wheat chromosome

DNA from the long arm of the wheat 5B chromosome was isolated from the complemented deletion line of the hexaploid 5Bdel wheat using microdissection of metaphase chromosomes. Isolated DNA was amplified by the primer-linker method. Size of the obtained DNA fragments varied in the range of 200 - 700 bp. The DNA probes obtained were shown by in situ hybridization to hybridize specifically with wheat chromosomes, and the radioactive label distribution was not uniform over the chromosomes
