Tissue culture of neem (Azadirachta indica A. Juss.) seedlings

Embryos, shoot tips, node segments and axillary buds of neem (Azadirachta indica A. Juss.) seedlings were cultured on modified Murashige and Skoog (MS 1962) medium containing 0, 0.5, 5, 10 and 50 micro mole BAP (6-benzylaminopurine) for 45 days. The medium containing 50 micro mole BAP gave the highest number of multiple shoots with 13.1 shoots/embryo. For shoot tip and node segment cultures, lower concentration of 5 micro mole BAP induced the highest shoot number with 6.5 and 6.9 shoots per shoot tip and node segment respectively. In the case of axillary bud cultures, 10 micro mole BAP gave the highest shoot number of 3.8 shoots. The formation of numerous shoots on the cotyledonous tissue was 16.3 % and 72.9 % when embryos were cultured in the medium containing 10 micro mole and 50 micro mole BAP respectively.
