Effect of glucose oxidase on the concentration of sucrose, glucose and fructose during the ensiling of ryegrass and alfalfa

Wilted grass Perun (Lolium multiflorum x Festuca pratensis, 2n=28) and alfalfa (Medicago sativa) were ensiled untreated (control) or treated with both the inoculant of Lactobacillus (Microsil) and the enzyme complex of cellulase, hemicellulase and glucose oxidase (GO) which contained the identical inoculant (Bactozyme). Saccharides were extracted with boiling ethanol and they were determined using ligand exchange chromatography on a strong cation exchanger in Na+ form. Ribose was used as an internal standard. Sucrose is most probably hydrolyzed with the original microflora, and the half-time of this reaction is approximately. 10 hours. The significant effect of GO was found in the initial stage of ensiling where glucose concentration was decreased by its oxidation to gluconic acid. The GO probably activated the original microflora which isomerized fructose to glucose within the heterofermentative stage of ensiling. The addition of glucose oxidase has the greatest effect in the initial hours of ensiling when glucose is rapidly oxidized to gluconic acid.
