Effects of light intensity on the morphogenesis of in vitro culturing leaf explant in African violets

The effects of light of various intensities (1000, 2000, 3000, 4000 and 5000 lx or darkness) on the morphogenesis of leaf blade explants cultured in the African violet (Saintpaulia ionantha H. wendl) cv. "Susie" were studied in vitro. The number of days required from leaf explant paced to callus and shoot formation was decreased at 4000 lx, but adventitious root formation was delayed at 5000 lx. The number and growth of proliferated shoots were not clearly affected by the light conditions. The number of formed adventitious root was increased and the growth was promoted at 4000 lx, but the former was decreased and the latter was hindered at 5000 lx. The fresh weight of calli, shoots and adventitious roots formed and the percentage of plants were increased with light intensity, but decreased in darkness or at 5000 lx. The results suggest that the morphogenesis of leaf explants of the African violets thus occurs optimally at 2000 - 4000 lx
