Antirheumatism activity of the total alkaloids from the stem of Makabuhay (Tinosphora rumphii, Boerl, Family Menispermaceae)

This study was undertaken to evaluate the alkaloid content from the stems of Makabuhay (Tinosphora rumphii, Boerl., family Menispermaceae) using chemical and instrumental techniques and to measure its anti-inflammatory activity. Positive results were obtained from the preliminary test on the makabuhay stems indicating the presence of alkaloids. Extraction of total alkaloids and partial purification yielded 0.043 percent and 0.091 percent. The alkaloidal extractive treated with alkaloid reagents proved the presence of alkaloid when it produced a white precipitate upon addition of Mayer's, Valser's and Wagner's and an orange-red precipitate with Dragendorff's reagent. The total number of alkaloids present in the alkaloidal extractive was determined by thin layer chromatography. Among the three solvent system used, solvent system for plate no. 2 gave the clearest presentation of the alkaloids spotted after spraying them with Dragendorff's spray agent. Rf value was calculated and recorded. Infrared spectrum yielded the following functional groups: alcohol, aromatic amine/C=NH, CH2, Ketone and benzene ring. The sample, semipurified alkaloids from makabuhay plant was administered orally in CK:RAR(SD) female rates. The result produced was a slight anti-inflammatory activity at 125 mg/kg giving 34.34 percent protection against edema formation. However, the 250 and 500 mg/kg doses did not produce any anti-inflammatory activity. The positive control showed 66.67 percent protection. This indicates that as the dosage increases the therapeutic activity of the alkaloidal extractive decreases
