Production and immobilization of a lipase from Rhizopus arrhizus Fischer from the synthesis of glycerides

A lipase from Rhizopus arrhizus was immobilized using silica gel as solid support. The kinetic parameters of the free and immobilized lipase showed Km and Vmax values of 0.0116, 0.0812 M and 0.004, 0.112 units/mg protein, respectively. Synthesis of glycerides was carried out using 6.2 lipase units of the immobilized lipase. ANOVA and CRD [completely randomized design] were employed to examine the effects of alcohol-oleic acid ratio, temperature, incubation time and moisture content on synthesis. Synthesis increased in a relatively straightforward manner in response to increase in temperature and incubation time and decrease in moisture content. Conditions were identified where the highest yield were obtained at 80 deg C, 96 hours incubation time, and 2.56 percent moisture content. The free evaporation method of removing moisture was found to be most effective compared with other methods in the experiment. Comparison of the effect of using lipase from different sources showed significant differences on the glycerides produced. Carica papaya latex lipase and lipozyme were able to synthesize both mono- and di-olein while immobilized lipase from R. arrhizus was able to synthesize only monoolein.
