Investigations on some aspects of in vitro culture of Annona cherimola Mill

The present study was carried out to investigate some aspects of in vitro culture of Annona cherimola Mill. Preconditioning of the mother plants by imposing etiolation pre-treatment, was found to have beneficial effect in increasing total shoot length, but not on multiple shoot proliferation produced by the cultured single-node shoot segment and shoot tip explants. Single-node shoot segment and axillary bud explants taken at different ages from Annona cherimola Mill. mother plants and cultured in media containing different concentration of cytokinins showed different growth patterns. The best growth observed for the single-node segment explants taken from 3-year-old seedlings was obtained in a medium without hormones, while for those taken from 9-year-old grafted tree obtained in medium containing 0.5 ppm BA + 0.5 ppm KN. In other experiment axillary buds taken from 3-year old seedlings showed the greatest percentage of burst buds and the longest shoots in a medium containing 3 ppm BA, while for those taken from the 9-year-old grafted tree in a medium containing 5 ppm BA. Different levels of combined KN and BA were tested, in order to study their effect on growth and the proliferation of shoot tip and axillary bud explants of Annona cherimola Mill. For the shoot tip and axillary buds the medium containing 0.5 ppm KN + 0.5 ppm BA secured the best conditions for proliferation. Increasing sucrose concentration in the medium was found to have a beneficial effect on increasing growth rate, up to 30 g/l, but when 60 g/l was used, a drastic reduction in growth rate was observed. Rooting of proliferating shoots in vitro was tested by using 50 mg/l IBA, but no success was obtained
