Karyological study of Abies concolor x Abies grandis calli and shoots regenerated in vitro

The extent of somaclonal variability in adventitious and axillary shoots regenerated in vitro from mature embryos and cotyledons of seedlings and in long-term callus cultures of vital hybrid A. concolor x A. grandis was determined. Karyological analysis was realised by determination of chromosome numbers in mitotic cells and by flow cytometric analysis of nuclear DNA content. Results showed that both adventitious and axillary shoots as well as long-term callus cultures are karyologically stable and remain at the diploid level.
