Species differences in establishing and morphology of the uterine tubal cell cultures

The aim of the work was to obtain long-term oviduct epithelian cell cultures of cow (BOEC), swine (POEC), rabbit (ROEC) and woman (HOEC) for further usage in co-culture systems. The techniques of cell isolation and species differences in cell morphology during culture were analyzed. The cells from bovine and rabbit oviducts were isolated by flushing with Dulbecco's PBS. The porcine and human cells were isolated by scraping of internal wall of the longitudinal cut oviducts. Five different culture media were used (Table 1). ROEC cells were less exacting as to medium composition and grew sufficiently in DME/F12, DME/F10 and Ham's F10 media. The monolayer with a prevalence of flat squamous epithelium cells was formed on 3rd day of culture. POEC and HOEC cells were very much more exacting as to culture conditions. Monolayers formed on the 8th day of culture and, in case of HOEC, it was rare and heterogenous (Table 2). The BOEC cells grew sufficiently in alpha-MEM or DMEM media and quickly formed a monolayer with a prevalence of the epithelial type cells. Optimal cell growth and gas change balances in the cultural environment were observed at 150 thsd BOEC cells in 1 ml of alpha-MEM medium.
