Model of foreign DNA transfer on porcine kidney epithelial cells

The aim of this work was to compare polybrene-mediate DNA transfer and electroporation in order to select more proper method for transfection of porcine cells. The porcine fetal kidney cell line (SPEV) and pSV2-neo plasmid with the marker gene Neo were used as a model. The cells were cultured in DMEM medium containing 10 % fetal calf serum. For polybrene method, the cells at monolayered state were treated without detaching from culture serface. The pSV2-neo plasmid was used at amount 2 mig ml** (-1). For electroporation, the cells were detached from culture dish by trypsin and were exposed to electric pulses of 15 misec duration in presence of 10 mig ml** (-1) of pSV2-neo.
