Plant disease control by using resistance is an ideal, simple, practical, less expensive to farmers. In order to obtain a resistant plant cultivar, reliable practical screening technique is needed. This research revealed the finding of the simple, reliable and practical field and laboratory screening techniques for Aspergillus flavus infection and aflatoxin contamination in corn. For field inoculation: the most effective and practical technique for large scale handling should be: injection of A. flavus suspension inoculum of 10** (7) conidia/ml concentration onto the silk channel of corn plant at three weeks after 100 % silking. For laboratory evaluation: Kernels inoculation by dipping in 10** (7) conidial suspension and then incubate in sterile petridish with two layers of filter paper on top of three layers of straw paper with only the second layer of straw which is moisted by soaking in water. This is the practical modified blotter method. These inoculation techniques have been used as routine work by Kasetsart University corn research in A. flavus, such as, screening for resistance to A. flavus infection and aflatoxin contamination in corn.
