Response of animals to vaccination against anthrax monitored by Elisa

Study is being undertaken to determine the antibody response as measured by ELISA of large and small ruminants to vaccination with a commercial Bacillus anthracis spore vaccine. Groups of sheep, goats, buffaloes and cattle were vaccinated with the recommended dose of vaccine and blood samples collected on a regular basis over the following year. An ELISA test was established using a washed cell preparation prepared from a solid agar culture of B. anthracis cells as the ELISA antigen. Polyacrylamide gel electrophoresis of the antigen showed the presence of a number of proteins including toxin components. Preliminary results from the test showed that all groups of animals responded to the vaccination however the antibody response of the sheep and goats was more marked than the response of the cattle and buffalo to the vaccination. Several cattle that received multiple doses of vaccine still produced a poor antibody response. Further work is underway to purify the ELISA antigen and to try and correlate the antibody response of the animals with protection to infection.
