Antennapedia variations in the E mutant group of Bombyx mori analyzed by southern blot hybridization

The E complex loci located on the 6th linkage group is known to contain homeotic genes determining the morphology of the abdominal segments of Bombyx mori. Another group of homeotic genes, including the B. mori Antennapedia (Bm Antp) gene, determine the identification of the thoracic segments. In the present study, 9 E mutant lines were analyzed by the Southern blot hybridization for restriction fragment length polymorphism (RFLP) using the Bm Antp gene sequences as probes. Of the mutants 4 showed the polymorphism corresponding to the segregation of the abdominal morphology. These results indicate the tight linkages between a part of the E loci and the Bm Antp gene. However, the crossing over between one of the E mutant genes (E-Nc) and the Bm Antp gene was observed at the recombination frequency of about 1.1%. This was reminiscent of the reported distance (1.4 cM) between the E loci and the Nc gene which contains the Bm Antp locus. Also banding polymorphism not related to the mutant character was seen suggesting that the Bm Antp locus has variable regions
